The answer would be Metaphase as during this stage chromosomes are fully condensed from chromatin and this stage is considered as the best to study the morphology of chromosomes and karyotypes are basically nothing but the arrangement of chromosomes in pairs in the descending order of their sizes.
Thus, Metaphase is best used for this purpose.
